چکیده
dITP may be generated from dATP by a slow, nonenzymatic hydrolysis. While [H]dITP was degraded rapidly to [H]deoxyinosine by HeLa cell nuclear extracts, no net degradation of [H]dITP was observed in the presence of physiological concentrations of ATP, apparently because the extract contained deoxynucleoside diphosphate kinase activity that regenerated [H]dITP from [H]dIDP. Isolated HeLa cell nuclei, as well as partially purified DNA polymerase a, incorporated [H]dITP into DNA at 50-60* of the rate of [H]dGTP incorporation. No rapid release of the Incorporated radioactivity was observed. The molecular weight of nascent DNA containing dIMP residues, however, decreased slightly after prolonged incubation 1n the presence of EDTA, suggesting that a repair process is initiated in dIMP-containing chromatin. Furthermore, release of free [H]hypoxanthine from [H]dIMP-contain1ng DNA was detected after incubation with nuclear extracts in the presence of EDTA, suggesting the presence of hypoxanthine-DNA glycosylase activity in HeLa cell nuclei.

Inhibition of ribonucleotide reductase, DNA synthesis, and L1210 leukemia by guanazole.
Guanazole (3,5-diamino-l,2,4-triazole; NSC 1895) was found to inhibit the incorporation of adenine, hypoxanthine, and uridine into DNA to a much greater extent than into RNA in LI210 leukemia cells in vivo. Thymidine incorporation into DNA was inhibited with concomitant accumulation of dTTP in LI 210 cells. Leucine incorporation into protein was not inhibited. The incorporation of formate-14C i...
متن کاملGnidilatimonoein from Daphne mucronata inhibits DNA synthesis in human cancer cell lines
The anticancer agents from plant sources usually exert their action through a wide range of mechanisms. As part of our studies of plants for new anticancer agents with emphasis on Thymelaeaceae family, we examined the cytotoxicity and anti-tumor activity of the water extract of D. mucronata leaves against induced breast tumor in rats. In the current study, we were interested to obtain some know...

متن کاملRegulation of de novo purine synthesis in human bone marrow mononuclear cells by hypoxanthine.
In previous studies from this laboratory, human bone marrow hypoxanthine concentrations were found to average 7.1 microM, three times higher than plasma hypoxanthine concentrations measured simultaneously. To assess the significance of this finding, the relationship between hypoxanthine concentration and the rate of purine nucleotide synthesis by the de novo pathway was studied in normal human ...
